Seven mammalian sirtuins have been identified so far (SIRT1 to SIRT7) 4,12. In the following years, there was further re-search on sirtuins, compounds modulating their activity, bioavailability, and their connection with metabolic functions (Figure 1). In 2003, David Sinclair showed that the activity of sirtuins could be modified by other substances, for example, resveratrol. Geneticist Amar Klar first described sirtuins in the 1970s by discovering the Sir-2 (Yeast Silent Information Regulators II, SIR2) gene in Saccharomyces cerevisiae cells. However, under adverse conditions, these pathways shift to protective and stress-bearing modes, eventually leading to an extended lifespan.
Including up to 127 covariates (based on ref. 34), e.g., assay center, dilution factors, blood draw time, and socioeconomic status indicators, or excluding related individuals from the analysis all showed negligible effects on the genetic findings (Supplementary Fig. 10). After the QC steps, our study included altogether 177,499 men and 205,141 women. However, twin studies indicate that heritability of serum T is relatively high in both sexes, being up to 65% in males19,20. Yet, these studies have yielded partly mixed results, and, in many instances, the proposed relationships between T, complex traits and disease remain elusive2,7–11.
Thus, there may be a therapeutic role for testosteronereplacement among menopausal and postmenopausal women, although careful assessment of themenopause age and monitoring of symptoms should be considered so that the lowest effectivedose of testosterone can be selected. Upon completion of menopause, average concentrations oftestosterone in women are approximately 15% of premenopause levels (54,55). In thisreview, we provide a broad overview of the physiology and role that androgens and estrogensplay in enhancing healthy aging and human longevity. In women, an accelerated loss of musclemass and strength occurs at an earlier age than in men (2–6), but life expectancy is higher inwomen compared with men (7). We further note that the emphasis of our study has been on adult T levels, whereas some of the effects of T might be organizational, acting during specific developmental windows. The male-specific total T PGS protected from stroke in men, whereas the same PGS had opposite effects on stroke risk in women. This in turn might partly promote the widespread sex differences across multiple traits, and the unique sex-specificity of genetics of T.
As epigenetic changes in the human body are reversible, these biomarkers are therefore useful for the identification and validation of anti-aging interventions . Sirt1 based downregulation of apoptosis and inflammation leads to the extension of lifespan contrary to that enhanced lipid metabolism and insulin sensitivity are main factors that increase the lifespan. The supplementation of precursor molecule of NAD+, NMN (nicotinamide mononucleotide) can reduce the DNA damage, heart failure, and effects of acute renal injury in a SIRT1-dependent manner 269,270. According to recent studies, p53 and p73 play crucial roles in reproduction 251,252. The e4 type have been reported to have short telomeres that have a role in life expectancy . There is evidence that the increase in human lifespan started about 1.8 million years ago, at those times the human populations were likely to be homozygous for the e4 allele . They also emphasized that the carriers of e4 alleles have higher cholesterol levels with greater chances of plaque formation in the arteries and enhanced risk of cardiovascular diseases, dementia, stroke and Alzheimer’s disease .
